β-Catenin independent, co-receptor independent, not well characterized WNT pathway

Assay establishment
Due to the fact that some recombinant WNT proteins are sticky and hard to handle, we are looking for a company with broad experiences in assay development in the field of β-Catenin independent WNT signaling. Your aim should be to develop an easy and precise “ready-to-use” competitive ligand binding assay. This assay must be suitable to screen ≤ 100 receptor binding antibodies for their property to compete with the natural ligand.

During the establishing process you have to show
• the specificity of WNT binding to its receptor
• a dose dependent increase of the read-out signal
• no/low unspecific binding of WNT to other irrelevant protein
• a suitable positive/negative control incl. a dose response curve
• the intra-assay and inter-assay precision

The assay can be based on all conventional detection methods e.g. FACS, micro plate readers, fluorescence, luminescence, absorbance etc.

Deliverables to customer:
All experimental results of the assay development process, including raw data and a detailed protocol/SOP shall be delivered to the customer. The final product should give us the opportunity to directly apply the assay in our lab.

Timelines:
• Delivery of final product beginning of April 2015
